目的:研究稳定转染hif-1α shRNA载体对辐射诱导B16细胞周期和凋亡变化的影响。方法:采用PI标记,流式细胞术检测细胞周期和细胞凋亡的变化。结果:不同剂量照射后,对照组和sihif-a组G2+M细胞随着剂量的增加均出现了明显增加(P<0.01),相同剂量下,sihif-a组G2+M细胞明显低于对照组(P<0.05,P<0.01),G0/G1和S细胞也出现了一定程度的降低,而细胞凋亡率有增高趋势。结论:转染hif-1α shRNA载体能够降低辐射诱导的G2+M阻滞,增加辐射诱导的细胞凋亡。
Objective: To investigate the effects of hif-1α shRNA vector stably transfected on the cell cycle and apoptosis of B16 cells induced by radiation. Methods: PI staining and flow cytometry were used to detect the changes of cell cycle and apoptosis. Results: The G2 + M cells in control group and sihif-a group increased significantly with the increase of dose (P <0.01). The G2 + M cells in sihif-a group were significantly lower than those in control group Group (P <0.05, P <0.01), G0 / G1 and S cells also showed a certain degree of reduction, while the rate of apoptosis increased. CONCLUSION: Transfection of hif-1α shRNA vector can reduce radiation-induced G2 + M arrest and increase radiation-induced apoptosis.
