目的观察参附配伍对心肌缺血血瘀证模型大鼠血液流变学和认知能力的影响。方法 SD大鼠60只,随机分为6组,每组10只,分别为空白对照组(空白组)、模型组、参附注射液腹腔注射组(注射组)、参附注射液口服组(口服组)、参附汤高剂量组(高剂量组)、参附汤低剂量组(低剂量组)。除空白组外,其余各组以寒凝加药物干预的方法建立心肌缺血血瘀证动物模型,给予相应药物干预。观察各组动物全血黏度、血细胞参数[包括平均红细胞体积(MCV)、红细胞分配宽度(RDW-CV),平均血小板体积(MPV)和血小板分配宽度(PDW)]、血沉等血液流变学指标,以及认知能力的影响。结果 1与空白对照组比较,模型组大鼠心电、血细胞参数及血液黏度检测结果差异均有统计学意义(P<0.05),表明药物加寒凝成功造成血瘀模型。2与模型组比较,各给药组心电ST振幅偏移减少,低、中、高切值显著降低,MCV、RDW-CV、MPV值显著降低,跳台实验的错误次数显著减少,差异均有统计学意义(P<0.05);注射组、口服组的PDW值、血沉显著降低(P<0.05);注射组、口服组和高剂量组的潜伏时间显著增加(P<0.05)。与注射组比较,低剂量组ST振幅、全血黏度、血细胞参数(MCV、RDW-CV、MPV值)、血液沉降率和跳台实验的错误次数差异均有统计学意义(P<0.05)。注射组、口服组各指标比较,无统计学差异(P>0.05),表明其药效相当。3血液流变学和行为学指标的典型相关性分析结果显示,寒凝血瘀动物的血流变性和认知能力存在相关性。结论参附配伍可改善模型动物血液流变学性状,同时提高血瘀动物的认知能力。
Objective To observe the effect of Shen-Fu-compatibility on the hemorheology and cognitive abilities in rats with myocardial ischemia-blood stasis syndrome. Results 1 Compared with the blank control group, there were significant differences in ECG, blood cell parameters and blood viscosity between the model group and the control group (P <0.05). 2 Compared with the model group, the ECG amplitude of ECG decreased, the values of low, middle and high cut significantly decreased, the MCV, RDW-CV and MPV decreased significantly, and the number of error of jumping test decreased significantly (P <0.05). The PDW and ESR of the injected group and the oral group were significantly decreased (P <0.05). The latency of the injection group, the oral group and the high-dose group were significantly increased (P <0.05). Compared with the injection group, ST amplitude, whole blood viscosity, blood cell parameters (MCV, RDW-CV, MPV), blood sedimentation rate and the number of error in jumping test in low dose group were significantly different (P <0.05). There was no significant difference between each group in injection group and oral group (P> 0.05), indicating that the efficacy was comparable. The results of canonical correlation analysis between hemorheology and behavioral indexes showed that there was a correlation between hemorheology and cognition ability of blood stasis and blood stasis animals. Conclusion Shen-Fu compatibility can improve the hemorheology of model animals and increase the cognitive ability of blood-stasis animals.
