为建立一种检测瘤内血管生成准确定量的新方法,采用抗CD34单克隆抗体标记实体肿瘤组织中的血管内皮细胞,运用流式细胞术进行定量分析;并采用免疫组化法定量瘤内微血管密度,比较两种方法检测值的相关性。结果显示流式细胞术实验组与对照组平均检测值(%)具有显著性差异(P<0.001);其相关性分析提示流式细胞术内皮细胞百分比与免疫组化法微血管计数值紧密相关(CD34,r=0.913,P<0.001;Ⅷ因子相关抗原,r=0.868,P<0.001)。以上可见,流式细胞术可用于准确、快速、定量地检测实体肿瘤组织中的血管内皮细胞。
To establish a new method for the accurate quantification of intratumoral angiogenesis, anti-CD34 monoclonal antibodies were used to label vascular endothelial cells in solid tumor tissues and flow cytometry was used for quantitative analysis.
