目的探讨经前舒颗粒对经前期综合征肝气郁证模型大鼠海马雌激素受体(ER)蛋白表达及与配体结合活性的影响。方法以情志刺激为主多因素造模法复制经前期综合征肝气郁证大鼠模型,使用中药复方经前舒颗粒进行干预治疗,采用免疫组织化学实验和放射性受体配体结合实验,分别检测了正常对照组、PMS肝气郁证组、经前舒干预组大鼠海马中的ER半定量表达水平和与E2的结合活性。结果与正常组相比,模型组大鼠海马CA1、CA3区ER蛋白表达水平显著升高(P<0.01,P<0.05),ER与配体的结合活性也显著增强(P<0.05,P<0.05);给药组的差异无统计学意义。结论大鼠海马中的雌激素受体表达量及与配体结合活性异常升高是经前期综合征肝气郁证的病机之一,也是调肝方药经前舒颗粒的可能作用靶点。
Objective To investigate the effect of Jingqianshu Granule on the expression of estrogen receptor (ER) protein and ligand binding activity in the hippocampus of rats with premenstrual syndrome of qi stagnation syndrome. Methods Based on emotional stimulation, a multifactorial model was used to replicate the rat model of premenstrual syndrome of liver qi stagnation. The Chinese medicine compound Jingqishu granule was used for intervention. Immunohistochemistry and radioactive receptor ligand binding The levels of ER semi-quantitative expression and the binding activity to E2 in hippocampus of rats in normal control group, PMSGGYG group and Jingqianshuang preconditioning group were detected. Results Compared with the normal group, the expression of ER protein in hippocampal CA1 and CA3 of model group was significantly increased (P <0.01, P <0.05), and the binding activity of ER with ligand was also significantly increased (P <0.05, P < 0.05). There was no significant difference in the medication group. Conclusion The expression of estrogen receptor in hippocampus and the abnormal increase of ligand binding activity in rat hippocampus are one of the pathogenesis of premenstrual syndrome liver qi stagnation.
