目的观察黄芪多糖对慢性阻塞性肺疾病(COPD)大鼠肺组织中羟脯氨酸和基质金属蛋白酶-9(MMP-9)的影响。方法采用香烟熏吸加气管内注入脂多糖(LPS)法建立大鼠COPD模型。黄芪多糖高、中、低3个浓度及强的松灌胃干预30 d。生化法检测肺组织中羟脯氨酸含量,RT-PCR法检测MMP-9基因表达水平,免疫组化法半定量检测MMP-9蛋白含量。结果模型组大鼠肺组织羟脯氨酸含量和MMP-9表达水平较正常对照组明显增加(P<0.01);黄芪多糖干预后,羟脯氨酸呈剂量依赖性明显减少(P<0.05,P<0.01),MMP-9基因及蛋白表达水平亦明显降低,以中剂量效果最明显。结论黄芪多糖可减少COPD大鼠肺组织羟脯氨酸的含量和MMP-9的表达,这种作用与改善COPD的肺损伤程度有一定关系。
Objective To observe the effects of astragalus polysaccharides on the expression of hydroxyproline and matrix metalloproteinase-9 (MMP-9) in the lungs of rats with chronic obstructive pulmonary disease (COPD). Methods The model of COPD was established by cigarette smoke suctioning and intratracheal injection of lipopolysaccharide (LPS). Astragalus polysaccharide in high, medium and low concentration of 3 and prednisone intragastric intervention 30 d. The content of hydroxyproline in lung tissue was detected by biochemical method, the expression of MMP-9 gene was detected by RT-PCR and the content of MMP-9 was detected by immunohistochemistry. Results The levels of hydroxyproline and the expression of MMP-9 in the lung tissue of the model group were significantly increased compared with those in the normal control group (P <0.01). The hydroxyproline significantly decreased in a dose-dependent manner (P <0.05, P <0.01), MMP-9 gene and protein expression levels were significantly reduced, the most obvious effect in the dose. Conclusion Astragalus polysaccharide can reduce the content of hydroxyproline and the expression of MMP-9 in the lung tissue of COPD rats, which may be related to the degree of lung injury in COPD.
