Siddha (Vatharasavangam VRV)是印度南方Tamil Nadn及其毗邻地区常用的药物,对高血糖家兔具有降血糖作用,用于治疗糖尿病。本文研究VRV对离体家兔胰岛细胞由葡萄糖浓度改变引起的胰岛素释放的影响以及酸性磷酸酶活性的变化,以了解药物的降血糖机制。家兔24只,均分为正常对照组、正常给药组、高血糖组和高血糖给药组。家兔禁食后静脉注射四氧嘧啶80mg/kg,可使血糖浓度升高至200~240mg/dl。VRV与蜂蜜混合后灌胃给药,每日1次,每次50mg/kg。8周后处死动物,取出胰脏,用胶原酶消化后分离获得胰岛细胞,充以95%O_2: 5%CO_2(v/v),与5ml葡萄糖(0.5mg/ml)培养液于37℃温育20min。此后,将每10个胰岛转移入1ml克林
Siddha (Vatharasavangam VRV) is a commonly used drug in Tamil Nadn and its adjoining areas in southern India and has a hypoglycemic effect on hyperglycemic rabbits for the treatment of diabetes. This study was to investigate the effect of VRV on insulin release induced by changes in glucose concentration in isolated rabbit islet cells and changes in acid phosphatase activity to understand the hypoglycemic mechanism of drugs. Twenty-four rabbits were equally divided into normal control group, normal administration group, hyperglycemic group, and hyperglycemic administration group. Rabbits after fasting intravenous injection of alloxan 80mg/kg, can increase blood glucose concentration to 200 ~ 240mg/dl. The VRV was mixed with honey and administered intragastrically once a day for 50 mg/kg each time. After 8 weeks, the animals were sacrificed and the pancreas was removed. The pancreatic islet cells were isolated by collagenase digestion and filled with 95% O 2 : 5% CO 2 (v/v) and incubated with 5 ml glucose (0.5 mg/ml) at 37°C. Breed 20min.
