目的 探讨肿瘤周围注射经TNF α基因转导的舌癌引流淋巴结淋巴细胞 (DNL)体内分布的特点。方法 在建立人舌癌原位移植模型基础上 ,用氚标胸腺嘧啶核苷 (3H TdR)标记转TNF基因的舌癌DNL后肿瘤周围注射 ,分别检测注射后 6h、1 2h、2 4h、48h、1 68h肿瘤组织及各脏器单位重量的放射活性 (衰变率 ,DPM)。结果 各重要脏器及血液中单位重量的TNF/DNL在不同的时间由高到低依次是 :1 2h瘤、脾、肝、肺 ,2 4、48、1 68h瘤、脾、肺、肝。瘤体单位重量DPM最高 ,平均是正常组织的 1 0 .9倍 ,是外周血的 1 2 .5倍。结论 肿瘤部位注射可提高肿瘤局部的TNF/DNL细胞量 ,可望发挥更好的抑瘤效果
Objective To investigate the distribution of tumor-infiltrating lymphatic lymphocytes (DNL) transduced by tumor necrosis factor-α (TNF-α) gene in vivo. Methods The human tongue cancer xenografts were established by orthotopic transplantation of tongue cancer. The tumors were injected with 3H TdR labeled DNG after transfection of DNG of tongue cancer. The expression of T lymphocytes was detected at 6h, 12h, 24h, 48h , Radioactive activity (decay rate, DPM) per unit weight of tumor tissues and organs at 1 68h. Results The major organs and blood per unit weight of TNF / DNL at different times from high to low are: 1 2h tumor, spleen, liver, lung, 2, 48, 68h tumor, spleen, lung and liver. The highest unit weight of tumor was DPM, which was 10.9 times of the normal tissue and 12.5 times of the peripheral blood. Conclusion The injection of tumor site can improve the local tumor cell TNF / DNL cells, is expected to play a better anti-tumor effect
