根据已报道的黄瓜花叶病毒外壳蛋白基因序列合成引物,经RT-PCR扩增并克隆了我国黄瓜花叶病毒赤豆分离物的外壳蛋白基因。序列分析表明,外壳蛋白基因全长657个核苷酸,编码218个氨基酸,其核苷酸序列与黄瓜花叶病毒亚组I的分离物有极高的同源性,达94%-95%,与亚组II的同源性仅为74%~75%。将该基因片段克隆到pGEMX-1载体中,转入E.coil后诱导表达,经Westernblot分析证明,该基因已在大肠杆菌中正确表达。
According to the reported sequences of cucumber mosaic virus coat protein genes, the coat protein genes of Cucumber Mosaic Virus red bean isolates were amplified and cloned by RT-PCR. Sequence analysis showed that the coat protein gene was 657 nucleotides in length and encoded 218 amino acids. The nucleotide sequence of the coat protein gene was highly homologous to the isolate of cucumber mosaic virus subgroup I, reaching 94% -95% , With subgroup II homology of only 74% to 75%. The gene fragment was cloned into pGEMX-1 vector, transferred to E. After induced by coil, the result of Western blot analysis showed that the gene was correctly expressed in E. coli.
