目的:探讨补肾中药天癸方对雄激素致不孕大鼠(ASR)促排卵的作用机制。方法:新生SD雌性大鼠随机分为3组,ASR模型组:9日龄SD雌性大鼠皮下注射丙酸睾丸酮;中药组:ASR模型在80日龄起每日灌服天癸方;对照组:9日龄SD雌性大鼠皮下注射中性茶油。在106日龄左右(动情前期),放免法测定血Δ4-雄烯二酮(Δ4-A)、总睾酮(TT)、游离睾酮(FT)、雌二醇(E2)、胰岛素(Ins)和C-肽(C-P),RT-PCR方法检测胰腺及卵巢雄激素受体(AR)mRNA表达水平,并观察中药治疗前后上述指标的变化。结果:ASR模型血Δ4-A、TT、FT、E2、Ins、C-P均显著高于对照组(P<0.05,P<0.01),胰腺、卵巢的AR mRNA表达水平明显升高(P<0.01),中药天癸方治疗后,排卵率为58%,各项血激素及胰腺AR mRNA表达水平明显下降,与对照组比较有显著性差异(P<0.05),卵巢AR mRNA水平与治疗前相比无明显改变。结论:ASR模型血雄激素水平升高,上调胰腺、卵巢的AR mRNA表达,引起高胰岛素血症和无排卵。天癸方可降低血雄激素和胰岛素,使胰腺AR mRNA表达水平下降,纠正ASR模型的内分泌-代谢失调状态,虽然卵巢的AR mRNA表达水平仍较高,但在正常的激素环境下仍出现了排卵。
Objective: To explore the mechanism of tonifying kidney invigorating traditional Chinese medicine on ovulation induced by androgen-sterilized rats (ASR). Methods: Newborn SD female rats were randomly divided into three groups: ASR model group: 9-day-old SD female rats were injected subcutaneously with testosterone propionate; Chinese medicine group: ASR model was given Tianqifang daily from 80 days old; 9-day-old SD female rats were injected subcutaneously with neutral tea oil. At 106 days of age (pre-estrus), serum levels of Δ4-androstenedione (Δ4-A), total testosterone (TT), free testosterone (FT), estradiol (E2), insulin and C-peptide (CP) and RT-PCR were used to detect the mRNA expression of androgen receptor (AR) in the pancreas and ovary. The changes of these indexes were observed before and after the treatment with Chinese medicine. Results: The levels of Δ4-A, TT, FT, E2, Ins and CP in ASR model group were significantly higher than those in control group (P <0.05, P <0.01) (P <0.05). The ovarian AR mRNA level was significantly lower than that before treatment (P <0.05), and the ovulation rate was 58% Significant change. CONCLUSION: The blood levels of androgens in ASR model are up-regulated, and the mRNA expression of AR in pancreas and ovary is increased, causing hyperinsulinemia and anovulation. Tiangui Decoction can lower blood androgens and insulin, reduce the expression of AR mRNA in the pancreas and correct the endocrine-metabolic imbalance of the ASR model. Ovary still displays ovulation even though the mRNA expression level of AR is still high in the ovary .
