目的 研究槲皮素在缺氧条件下对血管内皮细胞的保护作用。方法 体外培养人脐静脉内皮细胞(HUVEC) ,缺氧造成内皮细胞损伤 ,通过放免法和比色法测定培养上清中ET 1、NO的水平。结果 HUVEC缺氧4 8h后 ,NO含量下降 ,而ET 1含量明显增加 ,如在缺氧前用不同浓度槲皮素 (10、2 0、4 0、80 μmol/L)与HUVEC预孵 4 8h ,则可对抗缺氧引起的ET 1和NO水平的改变。结论 槲皮素能分别提高和降低血管内皮细胞在缺氧时ET 1和NO的水平 ,可能因此而产生保护作用。
Objective To study the protective effect of quercetin on vascular endothelial cells under hypoxic conditions. Methods Human umbilical vein endothelial cells (HUVECs) were cultured in vitro and endothelial cells were damaged by hypoxia. The levels of ET 1 and NO in culture supernatants were determined by radioimmunoassay and colorimetry. Results After 48 hours of HUVEC hypoxia, the NO content decreased, while the ET 1 content increased significantly. For example, pre-incubation with HUVEC with different concentrations of quercetin (10, 20, 40, 80 μmol/L) for 48 hours before hypoxia Can counteract the changes in ET 1 and NO levels caused by hypoxia. Conclusion Quercetin can increase and decrease the levels of ET 1 and NO in vascular endothelial cells during hypoxia, which may result in protective effects.
