离体大鼠比目鱼肌分别在含0~6μmol/L硝苯地平(NF)和14C-葡萄糖(14C-Glu)的KRB液中温育,然后用不含14C-Glu的KRB液冲洗,每5分钟一次,共75分钟,后25分钟用含10U/L胰岛素(Ins)的KRB液冲洗。结果Ins使14C-Glu的分段流量(λ)显著增高,说明Ins加速了葡萄糖在细胞内外的转运。NF浓度4μmol/L和6μmol/L两组在Ins负荷后,λ值显著低于0~2μmol/L各组,提示高浓度的NF抑制Ins介导的葡萄糖在肌细胞的转运。
Isolated rat soleus muscle was incubated in KRB solution containing 0-6 μmol / L Nifedipine (NF) and 14C-Glu (14C-Glu) and then rinsed with 14C-Glu-free KRB solution every 5 minutes Once for a total of 75 minutes, and 25 minutes later rinsed with KRB solution containing 10 U / L insulin (Ins). Results Ins significantly increased the fractional flow (λ) of 14C-Glu, indicating that Ins accelerates glucose transport both inside and outside the cell. The values of λ in 4μmol / L and 6μmol / L NF groups were significantly lower than those in 0 ~ 2μmol / L group after Ins load, suggesting that high concentration of NF inhibited Ins-mediated glucose transport in muscle cells.
