目的:研究胰腺癌中CXCL12-CXCR4生物学轴(CXCL12-CXCR4 biological axis)与血管内皮生长因子-C(vascular endothelial growth factor-C,VEGF-C)的表达情况,探讨其表达与胰腺癌的临床病理关系。方法:应用免疫组织化学和RT-PCR技术检测30例胰腺癌、癌旁、正常胰腺和胰周淋巴结组织中CXCL12、CXCR4和VEGF-C的表达情况。结果:CXCL12蛋白在胰腺癌组织中呈低表达状态,在正常胰腺组织、癌旁组织和胰周淋巴结组织均呈中等表达状态(P<0.01),RT-PCR结果也证实此结果,与胰腺癌组织中的表达相比较差异均有统计学意义(P<0.01)。CXCR4蛋白在胰腺癌组织、癌旁组织和胰周淋巴结均呈高表达状态,在正常胰腺组织中呈低表达状态,RT-PCR结果同样证实。经Spearman检验,胰腺癌组织中CXCR4与VEGF-C蛋白的表达具有显著的相关性(P=0.038)。结论:CXCR4的表达与胰腺癌淋巴结转移密切相关,其高表达对胰腺癌中微淋巴管的生成可能起促进作用。胰腺癌组织中CXCR4与VEGF-C蛋白的表达具有显著的相关性,两者可能对胰腺癌的淋巴管生成和淋巴结转移起协同作用。
OBJECTIVE: To study the expression of CXCL12-CXCR4 biological axis and vascular endothelial growth factor-C (VEGF-C) in pancreatic carcinoma and to investigate the relationship between the expression of CXCL12-CXCR4 biological axis and clinical manifestations of pancreatic cancer Pathological relationship. Methods: The expressions of CXCL12, CXCR4 and VEGF-C in 30 cases of pancreatic cancer, paracancer, normal pancreas and peritumoral lymph nodes were detected by immunohistochemistry and RT-PCR. Results: The expression of CXCL12 protein in pancreatic cancer tissues was low, and it was moderately expressed in normal pancreatic tissues, paracancerous tissues and peripancreatic lymph nodes (P <0.01). The results of RT-PCR also showed that CXCL12 was associated with pancreatic cancer There were significant differences in the expression in the tissues (P <0.01). The expression of CXCR4 protein in pancreatic cancer tissue, paracancerous tissues and peripancreatic lymph nodes were all highly expressed, which showed low expression in normal pancreatic tissues. The results of RT-PCR were also confirmed. The Spearman test showed that there was a significant correlation between the expression of CXCR4 and VEGF-C in pancreatic cancer (P = 0.038). Conclusion: The expression of CXCR4 is closely related to the lymph node metastasis of pancreatic cancer. The high expression of CXCR4 may promote lymphangiogenesis in pancreatic cancer. The expression of CXCR4 and VEGF-C protein in pancreatic cancer has a significant correlation, both of which may have a synergistic effect on lymphangiogenesis and lymph node metastasis of pancreatic cancer.
