以[Mo17(rf_3rf_3)CMS-唐徐×HZ_1(Rf_3Rf_3)N]×Mo17(rf_3rf_3)N回交群体作为基因定位群体。采用RFLP和RAPD分子标记技术,定位了Rf_3/rf_3基因。RFLP分析表明,Rf_3/rf_3基因位于第二染色体长臂上的UMC36A和UMC49分子标记之间,其遗传距离分别为12.7cM和4.8cM。采用BSA法,筛选了340个10mer随机引物,找到与Rf_3基因紧密连锁的分子标记RAPD Eo8-1.2,将Rf_3/rf_3基因的标记距离缩小到2.7cM,为Rf_3/rf_3基因辅助选择提供了有价值的分子标记。
The [Mo17 (rf_3rf_3) CMS-Tang Xu × HZ_1 (Rf_3Rf_3) N] × Mo17 (rf_3rf_3) N backcross population was used as the gene mapping population. The RFLP and RAPD markers were used to locate the Rf_3 / rf_3 gene. RFLP analysis showed that the Rf_3 / rf_3 gene was located between the UMC36A and UMC49 molecular markers on the long arm of the second chromosome with genetic distances of 12.7cM and 4.8cM, respectively. By using BSA method, 340 10mer random primers were screened to find RAPD Eo8-1.2, a marker closely linked to Rf_3 gene, and the marker distance of Rf_3 / rf_3 gene was reduced to 2.7cM, which provided valuable information for Rf_3 / rf_3 gene aided selection The molecular markers.
