目的:建立同时测定神农茶颗粒中夏佛塔苷、异夏佛塔苷、去氧地胆草内酯、4,5-二咖啡酰奎宁酸4个成分的高效液相色谱测定方法。方法:采用Hypersil C18色谱柱(200 mm×4.6 mm,5μm);以乙腈-0.025 mol·L-1磷酸溶液为流动相进行梯度洗脱,体积流量:1.3 ml·min-1,检测波长:270 nm(夏佛塔苷和异夏佛塔苷)、208 nm(去氧地胆草内酯)、327 nm(4,5-二咖啡酰奎宁酸),柱温为室温。结果:夏佛塔苷、异夏佛塔苷、去氧地胆草内酯和4,5-二咖啡酰奎宁酸分别在5.850~117.000,4.650~93.000,4.160~83.200,5.470~109.400μg·ml-1范围内线性良好,r值均大于0.999,平均加样回收率分别为97.70%、96.87%、97.53%、99.29%,RSD分别为1.40%、1.13%、1.69%、1.01%(n=6)。结论:该方法简便,结果准确,可用于神农茶颗粒的含量测定。
OBJECTIVE: To establish a method for the simultaneous determination of four components of siaftatidine, isoxifostidine, deoxycholic acid, and 4,5-dicaffeoylquinic acid in Shennongcha granules by high performance liquid chromatography. METHODS: Hypersil C18 column (200 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile-0.025 mol·L-1 phosphoric acid solution with gradient elution volume of 1.3 ml · min-1 and detection wavelength of 270 nm (asaftatidin and isofractaffioside), 208 nm (deoxynivalenolactone) and 327 nm (4,5-dicaffeoylquinic acid) at room temperature. Results: Clovetazin, isoflavoside, deoxycholic acid and 4,5-dicaffeoylquinic acid were in the range of 5.850 ~ 117.000, 4.650 ~ 93.000, 4.160 ~ 83.200 and 5.470 ~ 109.400μg · The average recoveries were 97.70%, 96.87%, 97.53% and 99.29%, respectively, with RSDs of 1.40%, 1.13%, 1.69% and 1.01% (n = 6). Conclusion: The method is simple and accurate and can be used to determine the content of Shennong tea granules.
