目的:研究重度肥胖病人淋巴细胞线粒体呼吸链的功能。方法:测定59例重度肥胖女性(肥胖组,体重指数为49.6±1.7,年龄为41.1±1.1岁)与60例正常体重女性(对照组,体重指数为22.5±0.6,年龄为44.1±1.6岁)的淋巴细胞线粒体呼吸链中第三对复合酶体-辅酶Q细胞色素c氧化还原酶(CⅢ)的活性及酶动力学分析。结果:肥胖组CⅢ的活性显著高于对照组(P<0.01),以Michaelis-Menton酶动力学方法分折,肥胖组CⅢ的最大反应速率(Vmax)及米氏常数(Km)均显著高于对照组(P<0.01)。结论:肥胖病人淋巴细胞线粒体呼吸链中电子流量增加伴复合酶体的效率下降,可能导致线粒体中自由基产生的增加.各种疾病的发病率增加。
Objective: To study the function of mitochondrial respiratory chain in patients with severe obesity. METHODS: Fifty-nine patients with severe obesity (obese, body mass index 49.6 ± 1.7, age 41.1 ± 1.1 years) and 60 normal-weight women (control group, body mass index 22.5 ± 0.6, age 44.1 ± 1.6 years old) in the mitochondrial respiratory chain of the third pair of complex enzyme-coenzyme Q cytochrome c oxidoreductase (C Ⅲ) activity and enzyme kinetic analysis. Results: The activity of CⅢ in obesity group was significantly higher than that in control group (P <0.01). The Michaelis-Menton kinetic method showed that the maximal response rate (Cmax) and the Michaelis constant (Km) Higher than the control group (P <0.01). CONCLUSIONS: Increased electron flux in the mitochondrial respiratory chain of obese patients with decreased efficiency of the complex enzymes may result in an increase in free radicals in the mitochondria. The incidence of various diseases increases.
