目的研究RNA解旋酶p68在真皮多能干细胞(dermal multipotent stem cells,DMSCs)增殖过程中的表达特点,并探讨其在创伤愈合中的意义。方法通过免疫细胞化学和Western blot方法检测p68在DMSCs不同生长状态的表达特点,早期伤口液刺激对DMSCs中p68表达的影响并通过MTT法检测DMSCs增殖的变化,进一步通过RNAi抑制p68表达之后检测细胞增殖的变化特点。结果增殖活跃的DMSCs中p68表达较强,饥饿以后,p68表达降低;伤后1 d伤口液可明显促进DMSCs中p68的表达及细胞增殖(P<0.01);RNAi抑制p68表达之后DMSCs增殖明显受抑(P<0.01)。结论RNA解旋酶p68参与DMSCs的增殖调控,创伤微环境在修复细胞的启动中可能发挥作用。
Objective To investigate the expression of RNA helicase p68 in proliferation of dermal multipotent stem cells (DMSCs), and to explore its significance in wound healing. Methods The expression of p68 in DMSCs was detected by immunocytochemistry and Western blot. The effect of early wound fluid stimulation on the expression of p68 in DMSCs was assayed. The proliferation of DMSCs was detected by MTT assay. The expression of p68 was detected by RNAi. Proliferation changes characteristics. Results The p68 expression was significantly increased in proliferating DMSCs and decreased after starvation. The wounds at 1 day after injury significantly promoted the expression of p68 and the cell proliferation in DMSCs (P <0.01). The proliferation of DMSCs was significantly inhibited after RNAi inhibited p68 expression (P <0.01). Conclusions RNA helicase p68 is involved in the regulation of proliferation of DMSCs. The injured microenvironment may play a role in the initiation of repairing cells.
