采用木瓜蛋白酶水解海洋生物泥螺的软体组织,进行多肽的抗肿瘤活性研究。通过单因素实验和正交实验来确定最佳酶解条件,采用MTT实验方法探索泥螺多肽的抗肿瘤活性。结果表明,当酶解条件为温度40℃、料液比1:1、pH6、酶解时间8 h、加酶量0.36%时,泥螺软体组织经木瓜蛋白酶水解制备所得的多肽的氨基氮含量最高(1.62 mg/mL),并且对肺癌A-549细胞和前列腺癌PC-3细胞的体外增殖表现出一定的抑制作用。
Papain was used to hydrolyze the soft tissues of marine mud snail to study the antitumor activity of the polypeptide. Through single factor experiments and orthogonal experiments to determine the optimal hydrolysis conditions, the MTT assay to explore the anti-tumor activity of the snail polypeptide. The results showed that when the enzymolysis temperature was 40 ℃, the ratio of material to liquid was 1: 1, pH6, enzymolysis time was 8 h and the amount of enzyme was 0.36%, the amino nitrogen content of Papilocephala spiralis tissue hydrolyzed by papain (1.62 mg / mL), and showed some inhibitory effects on the proliferation of A-549 lung cancer cells and PC-3 prostate cancer cells in vitro.
