【目的】观察不同剂量的三氧化二砷(As2O3)对成年大鼠生精细胞凋亡的影响。【方法】40只健康雄性SD大鼠随机分成对照组、低剂量组(0.0375g/L)、中剂量组(0.075g/L)、高剂量组(0.15g/L)4组,灌胃法连续给药16周后对各组大鼠睾丸取材进行精子头计数,并计算睾丸脏器系数、每日精子生成量(DSP)。采用甲基绿-派诺宁染色法和TUNEL法检测大鼠生精细胞凋亡情况。【结果】①中、高剂量组睾丸精子头计数和DSP均低于对照组(P<0.05);②与对照组相比,中、高剂量组生精细胞凋亡指数(AI)均显著升高(P<0.01);③生精细胞AI与DSP呈负相关(r=-0.563,P<0.01)。【结论】一定剂量As2O3可通过促进生精细胞凋亡而导致精子生成量的减少,从而产生雄性生殖毒性。
【Objective】 To observe the effects of different doses of arsenic trioxide (As2O3) on apoptosis of spermatogenic cells in adult rats. 【Methods】 Forty male SD rats were randomly divided into control group, low dose group (0.0375g / L), middle dose group (0.075g / L) and high dose group (0.15g / L) After 16 weeks of continuous administration, sperm head counts were taken from testis in each group, and testicular organ coefficient and daily sperm production (DSP) were calculated. The apoptosis of spermatogenic cells in rats was detected by Methyl Green - Pyronin staining and TUNEL method. 【Results】 ①The testicular sperm count and DSP in the medium and high dose groups were significantly lower than those in the control group (P <0.05); ② Compared with the control group, the spermatogenic apoptosis index (AI) (P <0.01). ③ AI of spermatogenic cells was negatively correlated with DSP (r = -0.563, P <0.01). 【Conclusion】 A certain dose of As2O3 can induce the spermatogenic cell apoptosis and lead to the reduction of sperm production, thus resulting in male reproductive toxicity.
