目的探讨异丙肾上腺素对失神经骨骼肌萎缩机制的防治情况。方法选取60只wista大鼠,制作成标准的腓肠肌失神经支配、右侧坐骨神经离断模型,随机分成观察组30只和对照组30只,应用Western Bloting和RT-PCR检测不同时间段蛋白质、mRNA、MuRF、1NF-JB的表达水平。结果两组在肌湿重维持率、NF-JBP65、MuRF1等方面相比差异均具有显著性(P<0.05)。结论异丙肾上腺素在抑制NF-JB/MuRF1通路中成效显著,能够达到防治失神经骨骼肌萎缩的目的。
Objective To investigate the prevention and treatment of isoproterenol on the mechanism of denervated skeletal muscle atrophy. Methods Sixty Wistar rats were selected and made into a standard denervated gastrocnemius and a right sciatic nerve sever model. The rats were randomly divided into observation group (30) and control group (30). Western Bloting and RT-PCR were used to detect the expression of protein and mRNA at different time points , MuRF, 1NF-JB expression levels. Results There was significant difference between the two groups in the maintenance of muscle wet weight, NF-JBP65 and MuRF1 (P <0.05). Conclusion Isoproterenol has a significant effect in inhibiting the NF-JB / MuRF1 pathway and can prevent and treat denervated skeletal muscle atrophy.
