目的寻找自然流产模型小鼠(CBA/J×DBA/2)子宫蜕膜组织与正常妊娠小鼠(CBA/J×BALB/c)子宫蜕膜组织之间差异表达的miRNAs。方法建立自然流产小鼠模型和正常妊娠小鼠模型,利用miRCURY LNATMmicroRNAArrays 10.0检测两组孕小鼠子宫蜕膜组织miRNAs的表达,筛选两组小鼠子宫蜕膜组织差异表达的miRNAs,并对其进行功能检索和靶基因预测。结果 CBA/J×DBA/2小鼠胚胎吸收率为30.9%,CBA/J×BALB/c小鼠胚胎吸收率为7.0%,两者差异有统计学意义(P<0.01)。miRNAs芯片数据表明流产蜕膜组织和正常蜕膜组织之间有13个miRNAs存在明显表达差异(ratios≥2.0或≤0.5),其中表达上调8个,表达下调5个。预测软件显示miR-21和miR-26a的靶基因分别是和妊娠相关的RECK和LIF两个因子。结论小鼠子宫蜕膜组织miR-21和miR-26a的差异性表达可能在小鼠胚胎自然流产过程中扮演重要角色。
Objective To investigate the differential expression of miRNAs between uterine decidual tissue of spontaneous abortion mouse (CBA / J × DBA / 2) and uterine deciduas of normal pregnant mice (CBA / J × BALB / c). Methods A spontaneous abortion mouse model and a normal pregnancy mouse model were established. MiRNAs were detected by miRCURY LNATM microRNA Arrays 10.0 in the uterine decidua tissues of two groups. The miRNAs differentially expressed in the decidual tissues of the mice were screened and their miRNAs Functional search and target gene prediction. Results The embryo absorption rate was 30.9% in CBA / J × DBA / 2 mice and 7.0% in CBA / J × BALB / c mice. The difference was statistically significant (P <0.01). miRNAs microarray data show that there are 13 miRNAs in aborted and normal decidua between abortion and abdomen. There are 8 miRNAs in the abortion group (n = 2.0 or ≤0.5), including up-regulated 8 and down-regulated 5. The prediction software showed that the target genes of miR-21 and miR-26a were RECK and LIF related to pregnancy respectively. Conclusion The differential expression of miR-21 and miR-26a in mouse uterine decidua may play an important role in the spontaneous abortion of mouse embryos.
