目的本文旨在探讨铅暴露对小鼠成神经瘤细胞(N2a)中突触囊泡蛋白C(synaptic vesicle protein C,SV2C)表达的影响,进一步了解铅对神经递质作用有关机制。方法 N2a细胞分为正常组、染铅Ⅰ组(10-4 mol/L醋酸铅),染铅Ⅱ(10-3mol/L醋酸铅)。分别在24和48 h进行RT-PCR和Western blot,观察不同组N2a细胞SV2C mRNA变化以及检测其蛋白表达水平。结果染铅Ⅰ、Ⅱ组N2a细胞中SV2C mRNA和蛋白表达水平均显著低于正常组,差异有统计学意义(P<0.05),但两剂量组之间无差异(P>0.05)。各组24和48 h比较,SV2C mRNA表达水平无差异(P>0.05),但染铅Ⅱ组SV2C蛋白表达水平48 h均明显低于24 h,有显著性差异(P<0.05)。结论本研究发现铅下调囊泡运输蛋白SV2C基因,减低了其蛋白水平,从而可能影响神经递质的分泌。
Objective To investigate the effect of lead exposure on the expression of synaptic vesicle protein C (SV2C) in mouse neuroblastoma cells (N2a) and to further understand the mechanism of lead on neurotransmitter. Methods N2a cells were divided into normal group, lead Ⅰ group (10-4 mol / L lead acetate), lead Ⅱ (10-3 mol / L lead acetate). RT-PCR and Western blot were performed at 24 and 48 h respectively to observe the changes of SV2C mRNA in different groups of N2a cells and to detect the protein expression. Results The levels of SV2C mRNA and protein in N2a cells were significantly lower than those in normal group (P <0.05), but there was no significant difference between the two groups (P> 0.05). Compared with 24 h and 48 h groups, there was no significant difference in SV2C mRNA expression (P> 0.05). However, SV2C protein expression levels in lead-exposed group at 48 h were significantly lower than 24 h (P <0.05). Conclusion This study found that lead down-regulated vesicular transport protein SV2C gene, reducing its protein level, which may affect neurotransmitter secretion.
