目的:建立高效液相色谱法测定岩黄连药材中脱氧卡维丁含量的方法,为完善质量评价提供依据。方法:Kromasil C_(18)柱(250 mm×4.6mm,5μm)为色谱柱,以乙腈-0.01 mol·L~(-1)磷酸二氢钾水溶液(调pH至5.0)(25:75)为流动相,流速1.0 ml·min~(-1),检测波长347 nm,柱温30℃。结果:脱氢卡维丁在0.1966~1.1796μg(r=0.999 7)范围内呈良好的线性关系,平均加样回收率为99.80%,RSD为1.35%。结论:所建立的方法简便、准确、重复性好。
Objective: To establish a method for determination of deoxycavidin in Rhizoma Rhizoma by high performance liquid chromatography and provide basis for improving quality assessment. Method: Kromasil C 18 column (250 mm × 4.6 mm, 5 μm) was used as a column, and acetonitrile-0.01 mol·L -1 potassium dihydrogen phosphate aqueous solution (adjusted to pH 5.0) (25:75) was used. Mobile phase, flow rate 1.0 ml·min~(-1), detection wavelength 347 nm, column temperature 30°C. RESULTS: The dehydrocavidin showed a good linear relationship in the range of 0.1966-1.1796 μg (r=0.999 7). The average recovery was 99.80% and the RSD was 1.35%. Conclusion: The established method is simple, accurate and reproducible.
