目的探讨Fas、P-糖蛋白(P-gp)、多药耐药1(mdr1)基因在急性白血病(AL)的表达及三者在多药耐药(MDR)中相关性的临床研究。方法对59例初发AL患者,应用流式细胞仪和半定量RT-PCR法分别检测治疗前后Fas和P-gp及mdr1表达情况。结果Fas在不同类型AL表达有差异;Fas+与P-gp+呈显著负相关,Fas+与mdr1+呈负相关,P-gp+与mdr1+呈显著正相关;COX分析结果显示:Fas和mdr1有判断预后价值;Fas+与Fas-组、P-gp+与P-gp-组、mdr1+与mdr1-组的CR率差异均有统计学意义。结论Fas高表达可能是AL预后良好因素;P-gp,mdr1高表达为预后不良重要因素。
Objective To investigate the expression of Fas, P-glycoprotein (P-gp), multidrug resistance 1 (mdr1) gene in acute leukemia (AL) and their correlation with multidrug resistance (MDR) Methods The expression of Fas, P-gp and mdr1 in 59 newly diagnosed AL patients were detected by flow cytometry and semi-quantitative RT-PCR respectively. Results There was a significant difference in the expression of Fas in different types of AL. There was a significant negative correlation between Fas + and P-gp +, negatively correlated with mdr1 +, and positive correlation between P-gp + and mdr1 +. COX analysis showed that Fas and mdr1 had prognostic value. The CR rates of Fas + and Fas-, P-gp + and P-gp- groups, mdr1 + and mdr1- groups were all significantly different. Conclusion The high expression of Fas may be a good prognostic factor for AL. The high expression of P-gp and mdr1 is an important prognostic factor.
