目的 用淡色库蚊溴氰菊酯抗性相关胰蛋白酶核酸探针检测淡色库蚊溴氰菊酯抗性。方法 用逆Northern鉴定抗药性品系和敏感品系中胰蛋白酶的表达差异 ,用随机引物法标记鉴定后的胰蛋白酶cDNA片段为探针Northernblot检测淡色库蚊的抗药性。结果 逆Northern显示胰蛋白酶在抗药性品系中的表达量是敏感品系的 4 33倍。以其为探针Northernblot检测显示 ,胰蛋白酶基因在抗药性品系中高表达 ,抗药性品系中胰蛋白酶的表达量是敏感品系的 3 90倍。结论 作为一种新的抗药性检测的分子生物学方法 ,胰蛋白酶核酸探针可用于区分抗药性品系和敏感品系
Objective To detect deltamethrin resistance in Culex pipiens pallens using the deltamethrin resistance-related trypsin-like nucleic acid probe of Culex. Methods Reverse Northern blot was used to identify the expression of trypsin in drug-resistant and susceptible strains. The cDNA fragments of the identified tryptic fragments were probed with Northern blot to detect the resistance of Culex pipiens pallens. Results Reverse Northern showed that the expression of trypsin in resistant strains was 433 times more sensitive than in susceptible strains. Northern blot analysis showed that trypsin gene was highly expressed in drug-resistant strains, and the expression of trypsin in drug-resistant strains was 3 90-fold higher than that in susceptible strain. Conclusion As a new molecular biology approach to drug resistance testing, trypsin nucleic acid probes can be used to distinguish drug-resistant and susceptible strains
