目的:探讨成年大鼠骨髓间充质干细胞(BMSCs)体外分化为神经前体细胞的能力。方法:原代:分离培养、鉴定;传代:在细胞因子和氧化剂作用下进行诱导。形态学观察,免疫细胞化学检测和流式细胞分析。结果:成功进行了BMSCs的原代和传代培养,诱导后有神经前体细胞产生,诱导后5小时平均分化率最高,约为49.79%。结论:BMSCs可进行体外分离、扩增,并能诱导分化为神经前体细胞。
Objective: To investigate the ability of adult rat bone marrow mesenchymal stem cells (BMSCs) to differentiate into neural progenitor cells in vitro. Methods: Primary: isolation and culture, identification; passage: induced by cytokines and oxidants. Morphological observation, immunocytochemistry detection and flow cytometry analysis. Results: Primary and subculture of BMSCs was successfully performed. After being induced, neural precursor cells were produced. The average rate of differentiation was the highest at 5 hours after induction, which was about 49.79%. Conclusion: BMSCs can be isolated and expanded in vitro and can differentiate into neural precursor cells.
