[目的]探讨八角科观赏植物组织培养及快繁技术体系。[方法]以八角科植物“孩儿莲”的顶芽为研究对象,以MS培养基为基本培养基,调查了培养基组成及抗褐化剂对顶芽茎尖存活和生长发育的影响。[结果]春季萌动初期的顶芽是八角科植物“孩儿莲”组织培养的最适外植体材料;灭菌条件以0.1%HgCl2溶液处理6min为宜,而常规酒精表面消毒会影响外植体存活;适宜的初代培养基为改良MS(改良大量元素与有机成分)+1倍量抗褐化剂+2.0mg/L6-BA+0.5mg/LNAA,继代培养基为改良MS(改良无机成分与有机成分)+1倍量抗褐化剂+2.0mg/L6-BA+0.1mg/LNAA。[结论]该研究结果为“孩儿莲”组培快繁技术的建立奠定了试验基础。
[Objective] The aim was to discuss the tissue culture and propagation technology system of Ornamental ornamental plants. [Method] With the top bud of plant “Brassica” as the research object and the MS medium as the basic medium, the effects of medium composition and anti-browning agent on the survival and growth of the shoot tip of the terminal bud were investigated . [Result] The top bud in the initial stage of spring germination was the most suitable explant material for the tissue culture of “Hibiscus”. The sterilization conditions were treated with 0.1% HgCl2 solution for 6 minutes, while the conventional alcohol surface disinfection would affect the outside The best primary culture medium was modified MS (modified macronutrients and organic components) plus 1 × browning agent + 2.0 mg / L 6-BA + 0.5 mg / L NAA, and the subculture medium was modified MS Inorganic components and organic components) +1 times browning agent + 2.0mg / L 6-BA + 0.1mg / LNAA. [Conclusion] The results of this study laid the experimental foundation for the establishment of tissue culture and propagation technology of “lotus lotus”.
