利用RAPD技术对枣 64个品种及类型的遗传变异进行了研究。从 12 0个 10bp随机引物中筛选出 4 0个多态性引物用于正式扩增 ,共扩增出 4 92条DNA带 ,其中多态性DNA带 2 93条 ,占 59.55% ,平均每个引物扩增的DNA带的数目为 12 .31条。有 13个品种及类型扩增出了 1个以上的特有RAPD标记 ,据此可以进行品种鉴定或早期性状预选。利用DNA扩增结果进行聚类分析 ,把供试的 64个品种及类型分为 8类 ,并对基本品种的确定及种下品种群的划分做了尝试。
The genetic variations of 64 varieties and types of jujube were studied using RAPD technique. Forty polymorphic primers were screened out from 12 0 random primers for amplifying 4 92 DNA bands, of which 2 93 polymorphic bands (59.55%) were detected, with an average of The number of DNA bands amplified by the primers was 12.31. Thirteen cultivars and types amplified more than one unique RAPD marker, allowing for the identification of cultivars or preselection of early traits. Based on the results of DNA amplification, the results of cluster analysis were used to classify the 64 cultivars and types tested into eight categories. At the same time, attempts were made to determine the basic varieties and to classify the cultivated species.
