目的 探讨用白细胞介素 2 (IL - 2 )激活的外周血 (APB)、脐带血 (ACB)和骨髓 (ABM)单个核细胞的培养上清对肿瘤细胞的作用 ,及其作用的可能机制。方法 应用 MTT法和集落培养法在体外检测了 APB、ACB和ABM培养上清对 HL 6 0和 K5 6 2细胞的作用 ,用 EL ISA法检测了各种上清中 TNF-α和 IFN-γ的产生情况。结果 APB、ACB和 ABM培养上清都可以产生 TNF-α和 IFN-γ,对 HL 6 0和 K 5 6 2细胞有一定抑制作用 ,以 APB培养上清最强。结论 应用 IL - 2净化自体干细胞移植物中混杂的肿瘤细胞时 ,培养上清中的细胞因子或其它成分也可以直接抑制肿瘤细胞。
Objective To investigate the effects of cultured supernatant of peripheral blood (PBMC), cord blood (ACB) and bone marrow (ABM) mononuclear cells activated by interleukin - 2 (IL - 2) on tumor cells and its possible mechanism. Methods The effects of APB, ACB and ABM culture supernatant on HL 60 and K5 6 2 cells were detected by MTT assay and colony culture assay. The levels of TNF-α and IFN-γ The production situation. Results The supernatant of APB, ACB and ABM could both produce TNF-α and IFN-γ, and inhibited the growth of HL-60 and K-562 cells. Conclusion When IL - 2 is used to purify mixed tumor cells in autologous stem cell grafts, the cytokines or other components in the culture supernatant can also directly inhibit the tumor cells.
