目的建立维血宁中虎杖苷、大黄素-8-O-β-D-葡萄糖苷、大黄素、大黄素甲醚4种化学成分的含量测定的HPLC方法。方法采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱;0.1%的磷酸溶液和乙腈进行梯度洗脱;柱温30℃;流速1 mL min-1;检测波长223 nm。结果在HPLC法中,虎杖苷、大黄素-8-O-β-D-葡萄糖苷、大黄素、大黄素甲醚的线性范围分别是:0.009~0.072 mg mL-1(r=0.999 1)、0.048~0.386 mg mL-1(r=1.000 0)、0.027~0.214 mg mL-1(r=1.000 0)、0.024~0.188 mg mL-1(r=1.000 0);平均回收率分别为98.8%、97.1%、95.5%、96.8%,RSD分别为0.6%、0.4%、0.3%、0.4%。结论所建立的方法专属性强,重复性好,可用于维血宁的质量控制。
OBJECTIVE To establish HPLC method for the determination of four chemical constituents of polydatin, emodin-8-O-β-D-glucoside, emodin and physcion. Methods Kromasil C18 column (4.6 mm × 250 mm, 5 μm) was used. The gradient elution was carried out with 0.1% phosphoric acid solution and acetonitrile. The column temperature was 30 ℃ and the flow rate was 1 mL min-1. The detection wavelength was 223 nm. Results The linear ranges of polydatin, emodin-8-O-β-D-glucoside, emodin and physcion were 0.009-0.072 mg mL-1 (r = 0.999 1) (R = 1.000 0) and 0.024-0.188 mg mL-1 (r = 1.000 0) in the range of 0.048 ~ 0.386 mg mL-1 and 0.027-0.214 mg mL-1 respectively.The average recoveries were 98.8% 97.1%, 95.5%, 96.8%, RSD 0.6%, 0.4%, 0.3%, 0.4% respectively.
