狗尾草(Setaria viridis是研究C4植物光合作用的优秀模型,同时也是研究非生物胁迫耐受性和生物能源原料的模型.经济、简单、灵活、快速、高效的转基因技术是模式植物应用发展的基础,本研究直接以狗尾草成熟种子为外植体,经过种子活化、金刚砂辅助农杆菌转化、抗性愈伤或丛生芽诱导、成苗、生根等基本过程,建立了一套新的狗尾草组织培养农杆菌转化技术,是国内外直接利用狗尾草活化种子进行农杆菌转化的初次报道.新的转化技术转化周期缩短为9~10周,转化效率A10达19.2%,ME34达22.2%,该方法对狗尾草品系的依赖性小,更适于难以利用已有方法建立转化体系的品系或野生种的转化.“,”Setaria viridis is an excellent model for studying the photosynthesis of C4 and is also a model for studying the tolerance of abiotic stress and bioenergy.Economical,simple,flexible,fast and efficient transgenic technology is the basis for the development of model plant applications.In this study,a new Setaria viridis tissue culture agrobacterium transformation technology was established by directly using mature seeds of Setaria viridis as explants,This is the first report for agrobacterium transfromation of Setaria viridis mature seeds at home and abroad.The system followed seed activation,emery-assisted agrobacterium transformation,resistance callus or multi-buds induction,seedling,rooting and other basic processes.The new transformation technology is shortened to 9 to 10 weeks,the transformation efficiency is up to 19.2% for A10 and 22.2% for ME34.The method was less dependent on the species of Setaria viridis and was more suitable for species which were difficult to use existing methods to establish a transformation system or wild species.
