目的:观察湿热痹片对灭活结核分枝杆菌(Mtb)诱导的佐剂性关节炎大鼠T细胞及细胞因子水平的影响。方法:采用热杀死结核分枝杆菌(H37Ra Mtb)免疫SD大鼠,以诱导佐剂性关节炎,用ELASA法检测大鼠外周血T淋巴细胞亚群、肿瘤坏死因子α(TNF-α)水平;同时测定大鼠踝关节滑膜组织中IL-1β,IL-6,IL-10及血管内皮细胞生长因子(VEGF)水平的变化。结果:与模型对照组比较,湿热痹组CD8+T细胞水平升高(P<0.05),CD4+/CD8+比值降低(P<0.05);促炎细胞因子TNF-α,IL-1β及IL-6水平下调(P<0.01),VEGF含量降低(P<0.01),同时抗炎因子IL-10水平上升(P<0.01)。结论:湿热痹片通过调节T细胞及细胞因子网络平衡而发挥对类风湿关节炎的治疗作用。
OBJECTIVE: To observe the effect of damp heat tablets on the inactivation of T cells and cytokines in MJ-induced adjuvant arthritis rats. METHODS: SD rats immunized with H37Ra Mtb were killed by heat to induce adjuvant arthritis, and peripheral blood T lymphocyte subsets and tumor necrosis factor-α (TNF-α) were detected by ELASA. The level of IL-1β, IL-6, IL-10 and the level of vascular endothelial growth factor (VEGF) in the synovium of the sacroiliac joint of rats were measured. RESULTS: Compared with the model control group, the CD8+ T cells in the Damp-Heat group increased (P<0.05), the ratio of CD4+/CD8+ decreased (P<0.05), and the pro-inflammatory cytokines TNF-α, IL-1β and IL-6. The levels were down-regulated (P<0.01), VEGF levels were decreased (P<0.01), and the levels of anti-inflammatory cytokines (IL-10) were increased (P<0.01). Conclusion: Damp-heating tablets can play a therapeutic role in rheumatoid arthritis by regulating the balance of T cells and cytokine networks.
