目的:探索新生大鼠脑皮质少突胶质Ⅱ型星形祖细胞(OP)的纯化及鉴定。方法:分离、培养新生大鼠皮质混合胶质细胞,采用改良的振荡分离纯化法获取OP细胞,并进行诱导分化和免疫组化鉴定。结果:用该方法获取的OP细胞A2B5和Nestin免疫染色为双阳性,细胞纯度可达90%以上,且具有分化为星形胶质细胞和少突胶质细胞的双分化潜能。结论:低速振荡结合2次接种可纯化得到高纯度的OP细胞,是一种简单、高效的OP细胞的纯化方法。
Objective: To explore the purification and identification of oligodendrocyte type Ⅱ astrocyte (OP) in the cortex of newborn rats. Methods: Newborn rat cortical mixed glia cells were isolated and cultured. OP cells were obtained by modified oscillatory separation and purification, and induced by differentiation and immunohistochemistry. Results: The immunofluorescence staining of A2B5 and Nestin obtained by this method was double positive. The purity of the cells was over 90%, and the potential of differentiation into astrocytes and oligodendrocytes was double differentiation. Conclusion: Low-speed shaking combined with 2 inoculation can be purified to obtain high-purity OP cells, which is a simple and efficient purification method of OP cells.
