目的探讨脑梗死患者外周血CD4+ CD28-T淋巴细胞亚群和血清中肿瘤坏死因子(TNF)-α在脑梗死发病中的作用及两者的相关关系。方法2006年2月至9月,在中国医科大学附属盛京医院神经内科运用免疫荧光标记技术和流式细胞仪分析以及双抗体夹心酶联免疫吸附试验(ELISA)法,对30例急性脑梗死患者(观察组)和30名健康志愿者(对照组)外周血CD4+ CD28-T淋巴细胞和血清中TNF-α质量浓度进行检测,并分析两者的相关性。结果急性脑梗死患者外周血中CD4+ CD28-T淋巴细胞和血清中TNF-α的质量浓度均比对照组显著增高,且两者具有相关性(r=0.51,P<0.05)。结论急性脑梗死时,患者外周血中CD4+ CD28-T淋巴细胞和血清中TNF-α的质量浓度均增高,两者相互作用,参与了急性脑梗死的异常炎性反应,在脑梗死发病中起重要作用。
Objective To investigate the role of CD4 + CD28-T lymphocyte subsets in peripheral blood and serum tumor necrosis factor-α (TNF-α) in the pathogenesis of cerebral infarction in patients with cerebral infarction and their relationship. Methods From February to September 2006, 30 patients with acute cerebral infarction were enrolled in Department of Neurology, Shengjing Hospital Affiliated to China Medical University. Immunofluorescence labeling, flow cytometry and double antibody sandwich enzyme-linked immunosorbent assay (ELISA) The levels of TNF-α in peripheral blood CD4 (superscript +) CD28 (superscript +) T-lymphocytes and serum in 30 healthy volunteers (control group) and 30 healthy volunteers (control group) were detected and their correlations were analyzed. Results The concentration of TNF-α in CD4 + CD28-T lymphocytes and serum in patients with acute cerebral infarction were significantly higher than those in control group (r = 0.51, P <0.05). Conclusions In patients with acute cerebral infarction, the levels of CD4 + CD28-T lymphocytes in peripheral blood and TNF-α in serum are increased, and the two interact with each other and participate in the abnormal inflammatory reaction of acute cerebral infarction.
