目的:研究聚酰胺富集木蝴蝶总黄酮的工艺参数及对阿尔兹海默病(AD)小鼠认知能力的影响。方法:以总黄酮收率为考察指标,经聚酰胺吸附柱富集后,采用紫外分光光度法测定总黄酮含量;腹腔注射东莨菪碱(3mg/kg)建造AD小鼠模型,木蝴蝶高、中、低剂量组分别给予1.786、3.572和5.358g/kg,空白对照组、模型组给予等体积的0.9%氯化钠溶液灌胃,每日2次,Morris水迷宫检测各组小鼠隐蔽平台实验潜伏期。结果:纯化工艺条件为:聚酰胺树脂(60-100)目,上样量3m L(60mg/m L),树脂径高比为1∶8,除杂剂10%乙醇,除杂剂体积12倍量柱体积,洗脱剂80%乙醇,洗脱速度1.0m L/min。与空白对照组同期比较,模型组小鼠各时段潜伏期显著延长(P<0.05);与模型组同期比较,高剂量组自第22天起显著缩短(P<0.05),中剂量组第28天显著缩短(P<0.05)。结论:本法富集纯化木蝴蝶总黄酮工艺稳定可行,木蝴蝶对AD小鼠空间学习记忆能力有一定的改善作用。
OBJECTIVE: To study the technological parameters of total flavonoids from Polygonatum mongolicum and its effect on cognition in mice with Alzheimer’s disease (AD). Conclusion: The enrichment and purification of total flavonoids from butterflies in Butterflies can improve the spatial learning and memory abilities of AD mice.
