目的:探讨prostasin基因在卵巢癌组织中的表达及意义。方法:①应用4 097靶基因点基因芯片对5例卵巢浆液性腺癌及5例正常卵巢组织进行基因表达谱分析,筛查差异表达基因。②应用RT-PCR检测10例卵巢癌组织、10例卵巢瘤组织及10例正常卵巢组织中prostasin mRNA的表达。结果:①基因芯片筛查出卵巢浆液性腺癌差异表达基因58个,其中表达增高(上调趋势)基因30个,表达降低(下调趋势)基因28个;prostasin基因为表达上调基因(cy5/cy3=3.018)。②prostasin基因在卵巢癌组织中表达显著增高(prostasin/actin:0.415±0.177),与正常卵巢组(prostasin/actin:0.192±40.128)及卵巢瘤组(prostasin/actin:0.237±0.232)比较无显著性差异(P>0.05)。结论:基因芯片可以用于筛查卵巢癌差异表达基因,prosta-sin为表达上调基因,prostasin mRNA在卵巢癌组织中表达显著高于在正常卵巢及卵巢瘤组织中表达。
Objective: To investigate the expression of prostasin gene in ovarian cancer and its significance. Methods: ① Gene expression profiles of 4 cases of ovarian serous adenocarcinoma and 5 cases of normal ovarian tissue were detected by 4 097 target gene microarray. The differentially expressed genes were screened. ② The mRNA expression of prostasin in 10 cases of ovarian cancer, 10 cases of ovarian tumor and 10 cases of normal ovarian tissue were detected by RT-PCR. RESULTS: ①Simultaneously, 58 differentially expressed genes in ovarian serous adenocarcinoma were screened by gene chip. Among them, 30 were up-regulated (down-regulated) and 28 were down-regulated (down-regulated); prostasin was upregulated (cy5 / cy3 = 3.018). (2) The expression ofprostasin gene was significantly increased in ovarian cancer tissues (prostasin / actin: 0.415 ± 0.177), which was not significantly different from that in normal ovarian tissue (prostasin / actin: 0.192 ± 40.128) and ovarian cancer group (prostacyclin / actin: 0.237 ± 0.232) Difference (P> 0.05). Conclusion: Gene chips can be used to screen differentially expressed genes in ovarian cancer. Prosta-sin is an up-regulated gene. Prostasin mRNA is significantly higher in ovarian cancer than in normal ovarian and ovarian tissues.
