目的建立HPLC同时测定鸡眼草药材中染料木素和山柰酚含量的方法。方法采用RPHPLC法。色谱柱为Diamons C_(18)(150 mm×4.6 mm,5μm),流动相为甲醇-体积分数0.1%磷酸水溶液(体积比为72∶28),流速为1.0 m L/min,检测波长为370 nm,柱温为30℃。结果染料木素和山柰酚质量浓度分别在0.50~4.02 mg/L和0.15~1.20 mg/L内与峰面积呈良好的线性关系,平均回收率分别为99.7%和100.9%,RSD分别为0.1%和0.7%(n=6)。结论建立的方法简便,准确,重现性好,可用于鸡眼草药材中染料木素和山柰酚的含量测定。
Objective To establish a method for the simultaneous determination of genistein and kaempferol in Cornice herb by HPLC. Methods Using RPHPLC method. The column was Diamons C 18 (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol-0.1% phosphoric acid solution (volume ratio 72:28), the flow rate was 1.0 m L / min and the detection wavelength was 370 nm, the column temperature is 30 ℃. Results The mass concentrations of genistein and kaempferol showed a good linear relationship with the peak area in the range of 0.50 ~ 4.02 mg / L and 0.15 ~ 1.20 mg / L, with the average recoveries of 99.7% and 100.9%, respectively, with RSDs of 0.1 % And 0.7% (n = 6). Conclusion The established method is simple, accurate and reproducible. It can be used for the determination of genistein and kaempferol in Cornopsis officinalis.
