目的:建立高效液相色谱同时测定人血浆中呱氨托美丁活性代谢物托美丁和MED5的方法。方法:血浆样品用50%的高氯酸沉淀,内标法定量。采用依利特C_(18)色谱柱(200 mm×4.6 mm,5μm),流动相为乙腈-0.05 mmol·L~(-1)KH_2PO_4(46:54),稀磷酸调节缓冲盐pH至3.50,流速1 ml·min~(-1),紫外检测波长为313 nm。结果:托美丁和MED5分别在0.02~4.05、0.02~0.99μg·ml~(-1)内线性良好(r>0.999),最低检测浓度均为0.02μg·ml~(-1),2个化合物的绝对回收率均大于80%,日间和日内RSD皆小于10%。结论:本方法操作简单,灵敏准确,稳定性好,适用于呱氨托美丁临床药物监测和药动学等的研究。
Objective: To establish a method for simultaneous determination of tolmetin and MED5 in human plasma by HPLC. Methods: Plasma samples were precipitated with 50% perchloric acid and quantified by internal standard method. The elution C_ (18) column (200 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-0.05 mmol·L -1 KH_2PO_4 (46:54). The dilute phosphoric acid was used to adjust the buffer pH to 3.50. 1 ml · min ~ (-1), UV detection wavelength of 313 nm. Results: Toludin and MED5 showed good linearity (r> 0.999) in 0.02 ~ 4.05 and 0.02 ~ 0.99μg · ml ~ (-1) respectively. The lowest detection concentrations were 0.02μg · ml ~ (-1) The absolute recoveries of all compounds were above 80% with RSD less than 10% during the day and the day. Conclusion: The method is simple, sensitive and accurate, and has good stability. It is suitable for the study of clinical drug monitoring and pharmacokinetics of ketamine.
