目的:研究益气养血泄浊法对慢性肾功能衰竭(慢性肾衰)大鼠肾组织形态的影响及血管内皮生长因子(VEGF)表达的作用机制。方法:建立5/6(Ablation/Infarction,A/I)肾切除慢性肾衰大鼠模型,设假手术组、模型组、西药(氯沙坦钾+福辛普利)组、中药(肾衰Ⅱ号方)组,干预后检测肌酐(Scr)、尿素氮(BUN)、VEGF蛋白、VEGF m RNA,观察肾组织形态;结果:治疗后与假手术组比较,两治疗组Scr、BUN明显下降(P<0.05,P<0.01);肾衰Ⅱ号方上调VEGF蛋白及VEGF m RNA、降低Scr、BUN方面优于西药组(P<0.05,P<0.01);两治疗组病理变化改善明显。结论:肾衰Ⅱ号方可改善慢性肾衰大鼠肾功能,减轻肾纤维化,上调VEGF表达,其抗肾间质纤维化作用可能与调节肾组织VEGF表达信号转道通路,调节肾小管周围毛细血管生成效应,改善肾间质缺血缺氧相关。
Objective: To study the effect of Yiqi Yangxue Xiezhuo Method on renal morphology and the expression of vascular endothelial growth factor (VEGF) in rats with chronic renal failure (chronic renal failure). Methods: A 5/6 (A / I) nephrectomy rat model of chronic renal failure was established. The rats in sham operation group, model group, western medicine (losartan potassium + fosinopril) Ⅱ group). After the intervention, creatinine (Scr), blood urea nitrogen (BUN), VEGF protein and VEGF m RNA were detected and the morphological changes of kidney were observed. Results: Scr and BUN of the two treatment groups were significantly decreased (P <0.05, P <0.01). Renal failure Ⅱ increased VEGF protein and VEGF mRNA expression, decreased Scr and BUN better than western medicine group (P <0.05, P <0.01). The pathological changes of two treatment groups improved obviously. Conclusion: Ren Shen Decoction Ⅱ can improve renal function, reduce renal fibrosis and up-regulate VEGF expression in rats with chronic renal failure, and its anti-renal interstitial fibrosis may be related to regulation of signal transduction pathway of VEGF expression in renal tissue, regulation of renal tubule Capillary angiogenesis effect, improve renal interstitial ischemia and hypoxia related.
