目的观察关节腔注射羧甲基壳聚糖(CMCTS)对骨关节炎(OA)模型中关节软骨退变及软骨中基质金属蛋白酶1,3(MMP1,3)mRNA表达的影响。方法16只大耳白兔行单侧前交叉韧带切断术,术后5周将动物随机分为2组,每组8只,实验组关节腔注射2%CMCTS0.3ml,每2周1次,共注射3次,对照组不注射任何药物。术后11周处死动物,比较各组股骨内髁关节软骨的大体变化,采用RTPCR方法检测软骨中MMP1和MMP3的mRNA表达。结果实验组软骨退变程度较对照组明显减轻,实验组软骨MMP1和MMP3的mRNA表达明显低于对照组(P<0.01)。结论关节腔注射CMCTS能够明显抑制OA软骨中MMP1和MMP3mRNA的表达,明显减轻软骨退变的程度,对软骨有一定保护作用。
Objective To observe the effect of CMCTS on the degeneration of articular cartilage and the expression of matrix metalloproteinase 1, 3 (MMP1, 3) mRNA in osteoarthritis (OA) model. Methods Sixteen rabbits were subjected to unilateral anterior cruciate ligament rupture. The rats were randomly divided into 2 groups (n = 8) at 5 weeks after operation. The experimental group received intraperitoneal injection of 2% CMCTS 0.3 ml every 2 weeks, A total of three injections, the control group did not inject any drugs. Animals were sacrificed 11 weeks after operation, and the gross changes of articular cartilage in the medial femoral condyle of each group were compared. The expressions of MMP1 and MMP3 mRNA in cartilage were detected by RTPCR. Results The cartilage degeneration in the experimental group was significantly reduced compared with that in the control group. The mRNA expression of MMP1 and MMP3 in the experimental group was significantly lower than that in the control group (P <0.01). Conclusion Intra-articular injection of CMCTS can significantly inhibit the expression of MMP1 and MMP3mRNA in OA cartilage, significantly reduce the degree of cartilage degeneration and have a certain protective effect on cartilage.
