以巨峰葡萄为材料,用浓度2%的单氰氨溶液涂抹芽眼促进休眠芽体提早萌发,利用cDNA-SRAP技术分析葡萄在单氰氨调控作用下基因差异表达情况,进而探讨单氰氨调控葡萄萌芽的分子机理。结果显示:在2.5%单氰氨处理下,葡萄基因表达明显。通过对差异片段回收、克隆、杂交鉴定和测序分析等研究,获得的一个已知基因功能差异表达的片段Esc1与玉米CAT2的同源性为74%,根据前人的报道这基因是与植物的抗逆性、生长、终止休眠等密切相关的因子,在单氰氨调控下差异表达。
Using Kyoho grapes as material, the buds were sprayed with 2% cyanamide solution to promote the early germination of dormant buds. The differential expression of grape under the regulation of cyanamide was analyzed by cDNA-SRAP, The molecular mechanism of budding. The results showed that under the treatment of 2.5% cyanamide, the expression of grape genes was obvious. The deduced amino acid sequence of Esc1 was 74% homologous to that of maize CAT2. According to the previous reports, this gene is homologous to plant Resistance, growth, termination of sleep and other closely related factors, under the regulation of cyanamide expression.
