目的观察体外 Hela 细胞在~(125)Ⅰ粒子照射下细胞凋亡和增殖的规律,探讨~(125)Ⅰ粒子对肿瘤的杀伤作用。方法用~(125)Ⅰ粒子照射 Hela 细胞,用不同的方法观察不同剂量和时间点细胞的增殖反应及形态变化。结果受照射细胞发生凋亡,于Ⅱb期出现凋亡小体;细胞凋亡率随累积剂量的增加而增加,于5 Gy 时出现凋亡率峰值;细胞克隆形成率显著下降,与凋亡形成呈反向关系。并有反剂量率效应。结论 ~(125)Ⅰ粒子对肿瘤的治疗作用可能为早期抑制肿瘤细胞增殖,晚期使肿瘤细胞大量凋亡。
Objective To observe the regularity of apoptosis and proliferation of Hela cells irradiated with ~ (125) Ⅰ particles in vitro and investigate the killing effect of ~ (125) Ⅰ particles on tumor cells. Methods Hela cells were irradiated with ~ (125) Ⅰ particles, and the proliferation and morphological changes of cells were observed by different methods at different dosage and time points. Results Apoptotic cells were found in irradiated cells. Apoptotic bodies appeared in stage Ⅱb. Apoptosis rate increased with the increase of accumulated dose. Peak of apoptosis occurred at 5 Gy. The rate of cell clone formation was significantly decreased, In reverse relationship. And anti-dose-rate effect. Conclusion The therapeutic effect of ~ (125) Ⅰ particles on tumor may be to inhibit the proliferation of tumor cells in early stage and to induce apoptosis of tumor cells in late stage.
