目的:建立了检测天花粉蛋白(Trichosanthin,TCS)肽图谱方法。方法:采用反相高效液相色谱(RP-HPLC)法和基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)法对天花粉蛋白的肽图谱进行了测定,比较了不同批次产品一级结构的一致性,并用MALDI-TOF-MS法进行了验证。结果:用RP-HPLC和MALDI-TOF-MS法检测天花粉蛋白胰酶裂解后的肽段,发现各批产品的一级结构具有一致性,此分析方法具有简单,灵敏度高,可靠等优点。结论:本法适应于天花粉蛋白药物的肽谱分析,同时也为分析PEG修饰天花粉蛋白药物肽图谱提供了可以借鉴定手段。
Objective: To establish a method for detecting peptide map of trichosanthin (TCS). Methods: The peptide map of trichosanthin was determined by reversed-phase high performance liquid chromatography (RP-HPLC) and matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) Structure consistency, and verified by MALDI-TOF-MS method. Results: The trichosanthin cleavage peptides were detected by RP-HPLC and MALDI-TOF-MS. The primary structure of each batch of products was found to be consistent. The assay was simple, sensitive and reliable. Conclusion: This method adapts to the peptide analysis of trichosanthin drugs, and also provides a reference for the analysis of PEG trichosanthin peptide mapping.
