为了研究SLE患者外周血淋巴细胞 (PBL)在体外培养下细胞凋亡指标Caspase 3和AnnexinV/PI的变化 ,使用AnnexinV联合PI染色和FITC标记的ActiveCaspase 3抗体染色 ,流式细胞术检测 4 4例SLE患者和 30例正常人外周血淋巴细胞在体外培养 4 8h后淋巴细胞 (PBL )凋亡的发生率。结果显示在体外培养作用下 ,活动期SLE患者PBL凋亡率较正常人显著增高(P <0 0 1) ,而静止期则无明显差别 (P >0 0 5 )。SLE疾病活动指数SLEDAI,抗dsDNA抗体与淋巴细胞Caspase 3和AnnexinV/PI表达呈明显的正相关 (P <0 0 5 )。活动期SLE患者PBL凋亡较正常人显著增高。淋巴细胞凋亡与SLE疾病活动指数之间具有明显的相关性 ,表明SLE存在体内凋亡或凋亡相关性免疫机制失调
In order to study the changes of apoptotic index Caspase 3 and Annexin V / PI in peripheral blood lymphocytes (PBL) of SLE patients in vitro, AnnexinV combined with PI staining and FITC-labeled ActiveCaspase 3 antibody staining were used to detect 44 cases The incidence of lymphocyte (PBL) apoptosis in SLE patients and 30 normal peripheral blood lymphocytes cultured in vitro 48 h. The results showed that the apoptosis rate of PBL in patients with active SLE was significantly higher than that in normal subjects (P <0.01), while there was no significant difference in resting state (P> 0.05). SLE disease activity index SLEDAI, anti-dsDNA antibody and lymphocyte Caspase 3 and AnnexinV / PI expression was positively correlated (P <0 05). Active SLE patients with PBL apoptosis was significantly higher than normal. Lymphocyte apoptosis and SLE disease activity index was significantly correlated, indicating that SLE exist in vivo apoptosis or apoptosis-related immune dysregulation
