目的:建立一种直接进样高效液相色谱法测定人血浆中的头孢唑林、头孢曲松、头孢唑肟的血药浓度。方法:选用浸透限制固定相,紫外检测器,使3种头孢菌素分别在流动相pH5.75、pH5.25和pH4.35条件下与内标及血浆中的蛋白质进行良好的分离。结果:制作了3种头孢菌素各自的标准曲线,测定方法的变异系数在1.2%~7.1%,回收率近100%。最后,实测了6例临床静脉输注头孢唑林、头孢曲松、头孢唑肟病人的血药浓度。结论:浸透限制固定相色谱法测定血浆中的头孢菌素,省去了前处理,快速简单,更适用于临床治疗药物监测。
Objective: To establish a direct injection of high performance liquid chromatography for the determination of plasma concentrations of cefazolin, ceftriaxone, and ceftizoxime in human plasma. Methods: The three kinds of cephalosporins were separated from the internal standard and the plasma proteins by mobile phase pH5.75, pH5.25 and pH4.35, respectively. Results: The standard curve of three kinds of cephalosporins was made. The coefficient of variation (CV) was between 1.2% and 7.1% and the recovery rate was nearly 100%. Finally, 6 cases of clinical intravenous cefazolin, ceftriaxone, ceftizoxime patients blood concentration. Conclusion: The determination of cephalosporins in blood plasma by the limit-of-penetration fixed-phase chromatography eliminates the need for pre-treatment and is quick and easy. It is more suitable for the monitoring of clinical therapeutic drugs.
