miR-92a-3p对3T3-L1前体脂肪细胞增殖与分化的影响

为探讨mi R-92a-3p对3T3-L1前体脂肪细胞增殖与分化的影响,运用脂质体转染化学修饰模拟物mi R-92a-3p mimic使mi R-92a-3p过表达,并采用实时荧光定量PCR、蛋白免疫印迹检测、油红染色、甘油三酯检测、CCK-8检测和细胞周期检测等方法,研究mi R-92a-3p对3T3-L1前体脂肪细胞增殖和分化为成熟脂肪细胞的影响。结果显示,mi R-92a-3p在3T3-L1前体脂肪细胞成脂分化过程中显著上调,过表达mi R-92a-3p能促进脂肪细胞分化指标基因(PPARγ、C/E B Pα、F ABP4)的m RNA和蛋白表达,增加甘油三酯的聚积,且能下调细胞周期G 1期标志基因(Cyclin D 1、D3和CDK4)的m RNA和蛋白表达。结果表明,mi R-92a-3p抑制3T3-L1前体脂肪细胞增殖并且促进其分化为成熟脂肪细胞。 To investigate the effect of mi R-92a-3p on the proliferation and differentiation of 3T3-L1 preadipocytes, mi R-92a-3p mimic was transfected with the chemical modification mimic to overexpress mi R-92a-3p The effects of mi R-92a-3p on the proliferation and differentiation of 3T3-L1 preadipocytes were studied by real-time fluorescence quantitative PCR, Western blotting, oil red staining, triglyceride assay, CCK-8 assay and cell cycle assay Mature adipocytes. The results showed that mi R-92a-3p significantly up-regulated the adipogenic differentiation of 3T3-L1 preadipocytes, and overexpression of mi R-92a-3p promoted the differentiation of adipocyte differentiation genes (PPARγ, C / EB Pα, F ABP4 ) M RNA and protein expression, increase the accumulation of triglycerides, and can down-regulate the cell cycle G 1-phase marker genes (Cyclin D 1, D3 and CDK4) m RNA and protein expression. The results show that mi R-92a-3p inhibits the proliferation of 3T3-L1 precursor adipocytes and promotes their differentiation into mature adipocytes.
