目的:探讨天花粉水煎剂在体外对人恶性黑色素瘤A-375细胞凋亡的影响。方法:应用DAPI荧光染色观察天花粉水煎剂作用后A-375细胞凋亡情况;采用流式细胞仪检测天花粉水煎剂对A-375凋亡率的影响;利用分光光度法检测不同质量浓度的天花粉水煎剂处理A-375细胞后,A-375细胞中Caspase-3蛋白活性情况。结果:与对照组相比,实验组荧光显微镜下可见典型凋亡小体,且呈时间和质量浓度依赖。同时,发现天花粉水煎剂可使A-375细胞凋亡率和Caspase-3活性明显升高,并呈时间和质量浓度依赖性。结论:天花粉有诱导人恶性黑色素瘤细胞A-375细胞凋亡的作用,其诱导黑色素瘤凋亡是通过影响凋亡因子Caspase-3蛋白活性途径实现的。
Objective: To investigate the effects of TCS decoction on the apoptosis of human malignant melanoma A-375 cells in vitro. Methods: Apoptosis of A-375 cells was observed by DAPI staining. Apoptosis rate of A-375 cells was detected by flow cytometry. Apoptosis rates of A-375 cells were measured by spectrophotometry Caspase-3 protein activity in A-375 cells treated with TCS treated A-375 cells. Results: Compared with the control group, typical apoptotic bodies were observed under the fluorescence microscope in the experimental group, and showed time and mass concentration dependence. At the same time, it was found that TCS could significantly increase the apoptosis rate and Caspase-3 activity of A-375 cells in a time-and mass-dependent manner. Conclusion: TCS can induce the apoptosis of human malignant melanoma cell line A-375, which induces the apoptosis of melanoma by affecting the activity of Caspase-3.
