目的应用高效液相色谱法(HPLC)建立黄海葵的HPLC指纹图谱,为其质量控制和药材鉴别提供依据。方法采用反相C18色谱柱,以乙腈-0.1%(V/V)磷酸水为流动相,二元线性梯度洗脱,流速为1.0 mL.min-1,柱温为室温,检测波长为210 nm,进样量5μL;采用高效液相色谱-质谱联用技术(HPLC-MS)对其特征峰进行快速分析;将相似度计算法用于8批黄海葵药材的评价。结果以7个共有峰为评价指标,建立了黄海葵药材的HPLC指纹图谱,通过相似度计算发现8批黄海葵相似度均在0.97以上;HPLC-MS分析获得的4个特征峰的质谱信息,可实现黄海葵药材指纹图谱特征峰的准确定位。结论该方法准确、简单、稳定性好,所建立的黄海葵药材的HPLC指纹图谱可作为药材定性真伪鉴别和质量控制的有效手段。
OBJECTIVE To establish HPLC fingerprinting HPLC for HPLC to provide a basis for quality control and identification of herbs. Methods The reversed-phase C18 column was used with mobile phase of acetonitrile-0.1% (V / V) phosphoric acid as mobile phase with a linear gradient of 1.0 mL · min-1. The column temperature was at room temperature and the detection wavelength was 210 nm , The injection volume was 5μL. The characteristic peaks were analyzed by HPLC-MS. The similarity calculation method was applied to the evaluation of eight batches of Herba Houttuyniae. Results Seven common peaks were used as the evaluation index to establish the HPLC fingerprints of Huanghai Kui medicinal materials. The similarity between the eight batches of Haemonchus anguillicaudatus was above 0.97. The mass spectral information of the four characteristic peaks obtained by HPLC- It can achieve the accurate positioning of the characteristic peaks of the fingerprints of Herba yangkuigensis. Conclusion The method is accurate, simple and stable. The established HPLC fingerprints of Herba Houttuyniae can be used as an effective means to identify the authenticity and quality control of medicinal herbs.
