目的探讨低剂量镉对雄性SD大鼠抗氧化损伤能力适应性反应的影响。方法将40只雄性SD大鼠随机分为4组,每组10只。对照组,每天皮下注射生理盐水1次,连续3d;0.3mg/kg染镉组,以0.3mg/kg的剂量(以Cd2+计)连续皮下注射镉3d;0.3mg/kg+5.0mg/kg染镉组,以0.3mg/kg的剂量连续皮下注射镉3d,d3注射镉8h后,再以5.0mg/kg剂量皮下注射镉1次;5.0mg/kg染镉组,前两天每天皮下注射生理盐水1次,d3与0.3mg/kg+5.0mg/kg染镉组同时皮下注射5.0mg/kg的镉。末次染毒后24h,以20%的乌拉坦1ml/kg麻醉动物,股动脉采血后,解剖动物,取肝脏;以邻苯三酚自氧化法分别测定血清和肝脏中SOD活性,以荧光光度法分别测定血清和肝脏中MDA的含量。结果 5.0mg/kg染镉组SOD活性低于0.3mg/kg+5.0mg/kg染镉组,差异有统计学意义(P﹤0.05);5.0mg/kg染镉组MDA含量高于0.3mg/kg+5.0mg/kg染镉组,差异有统计学意义(P﹤0.05)。结论皮下注射0.3mg/kg的镉可致雄性SD大鼠产生抗氧化损伤能力的适应性反应。
Objective To investigate the effect of low dose cadmium on the adaptive response to oxidative damage in male SD rats. Methods Forty male Sprague-Dawley rats were randomly divided into 4 groups with 10 rats in each group. The rats in the control group were injected subcutaneously with normal saline once a day for 3 consecutive days. 0.3 mg / kg cadmium exposure group was injected subcutaneously with 0.3 mg / kg cadmium (0.3 mg / kg) and cadmium (0.3 mg / kg) Cadmium group. The mice were injected subcutaneously with 0.3 mg / kg cadmium for 3 days and cadmium d3 for 8 hours. The rats were injected subcutaneously with 5.0 mg / kg cadmium once. Saline once, d3 and 0.3mg / kg + 5.0mg / kg cadmium group while subcutaneous injection of 5.0mg / kg of cadmium. The animals were anesthetized with 20% of urethane 1ml / kg 24h after the last exposure, and the animals were sacrificed after the blood was collected from the femoral artery. The activity of SOD in serum and liver were determined by pyrogallol autoxidation method. The contents of MDA in serum and liver were measured respectively. Results The SOD activity in 5.0mg / kg cadmium group was lower than that in 0.3mg / kg + 5.0mg / kg cadmium group (P <0.05). The content of MDA in 5.0mg / kg cadmium group was higher than 0.3mg / kg + 5.0mg / kg cadmium group, the difference was statistically significant (P <0.05). Conclusion Subcutaneous injection of cadmium at 0.3 mg / kg can induce adaptive responses to oxidative damage in male SD rats.
