目的:研究大孔树脂法分离纯化菊明降压有效部位总黄酮的工艺条件及参数。方法:以总黄酮和蒙花苷为考察指标,考察5种大孔树脂的吸附分离性能,筛选合适的大孔树脂型号,优化大孔树脂分离纯化的工艺条件和参数。结果:AB-8型树脂对菊明总黄酮和蒙花苷的吸附分离性能较好,最优工艺条件为上样浓度为生药0.25g.mL-1,上样量为2倍柱体积(BV),树脂径高比为1∶15,先用3BV纯化水洗脱除去杂质,再用80%乙醇3BV洗脱。结论:AB-8型大孔树脂在所确定的优化工艺条件下,可较好地吸附分离菊明总黄酮。
OBJECTIVE: To study the conditions and parameters of separation and purification of total flavonoids from buckwheat effective parts by macroporous resin. Methods: Total flavonoids and mandarin glycosides were investigated. The adsorption and separation performance of five kinds of macroporous resins were investigated. The suitable macroporous resin models were screened to optimize the technological conditions and parameters for the separation and purification of macroporous resins. Results: The AB-8 resin had better adsorption and separation performance on the total flavonoids and the monolinosides. The optimum conditions were as follows: the crude drug concentration was 0.25g.mL-1, the sample volume was 2 times the column volume (BV ), The resin diameter to height ratio of 1:15, first with 3BV purified water to remove impurities, and then 80% ethanol 3BV eluting. CONCLUSION: The AB-8 macroporous resin can adsorb and purify total flavonoids of kyriva under optimum conditions.
