目的:探讨葛根素对去卵巢大鼠T淋巴细胞亚群表达的影响。方法:通过不同剂量葛根素、倍美力溶液灌服去卵巢大鼠30天后,流式细胞术分析各组大鼠外周血T淋巴细胞亚群CD3+、CD4+、CD8+及CD4+/CD8+的表达。结果:给药30天后,模型组与正常组比较CD3+、CD4+、CD4+/CD8+表达明显下降(P<0.01),葛根素高剂量组、低剂量组与阴性对照组比较CD3+、CD4+、CD4+/CD8+的表达明显增加(P<0.05)。各组大鼠外周血T淋巴细胞CD8+表达无明显差异。结论:大鼠去势后外周血CD3+、CD4+、及CD4+/CD8+表达下降,葛根素能上调去卵巢大鼠外周血CD3+、CD4+及CD4+/CD8+表达,具有调节去卵巢大鼠免疫的作用。
Objective: To investigate the effect of puerarin on the expression of T lymphocyte subsets in ovariectomized rats. METHODS: After 30 days of ovariectomized rats were fed with different doses of puerarin and premarin solution, the expression of CD3+, CD4+, CD8+, and CD4+/CD8+ T lymphocyte subsets in peripheral blood was analyzed by flow cytometry. Results: After 30 days of administration, the expression of CD3+, CD4+ and CD4+/CD8+ was significantly decreased in the model group compared with the normal group (P<0.01). The CD3+, CD4+, and CD4+/CD8+ were compared between the high-dose and low-dose groups of puerarin and the negative control group. The expression was significantly increased (P<0.05). There was no significant difference in CD8+ expression of T lymphocytes in peripheral blood of rats in each group. Conclusion: The expression of CD3+, CD4+, and CD4+/CD8+ in the peripheral blood of ovariectomized rats decreased. Puerarin can up-regulate the expression of CD3+, CD4+, and CD4+/CD8+ in the peripheral blood of ovariectomized rats, and it can regulate the immune response of ovariectomized rats.
